主要功能用途

Leica DMi8是一款采用先进模块化设计，具有开放架构、可灵活配置各种光学应用模块的研究级高端倒置荧光显微镜，与徕卡Leica Application Suite X (LAS X) 成像软件相结合，既能完成常规细胞明场、相差对比、微分干涉对比(DIC)、调制对比（IMC）和荧光对比方法观察与成像，又可用于光转化、光消融、钙离子荧光等宽视场高速多通道高级荧光应用研究。

 

性能特点

DMi8多功能倒置荧光显微镜在光学接口设置、延时程序自动聚焦、多色荧光信号同步采集、模块化组合配置等方面的引入创新设计，特别适合活细胞成像应用。

1、智能化控制

可通过触摸屏、功能控制元件或PC软件实现完全智能自动化和操作所有显微镜功能。

机身前部可选配倾角可调大尺寸彩色触摸屏，为所有电动和编码功能的控制单元。操作界面友好直观，可控制和查看显微镜各项工作状态：观察方法、物镜放大倍数及物镜类型（干镜、水镜、油镜），光源类型和光照强度，控制视场光阑及孔径光阑、激发光滤块转换等。

当选择特定的对比度模式，DMi8A显微镜可自动将所有组件移到光路中，将照明设置、对焦、光照强度和孔径和视场光阑调整到该模式。

 

2、多Infinity Port光路接口

DMi8 显微镜光学扩展模块含有两个新型无限远光路接口（Infinity Port）。主机的左侧、右侧、底部及目镜筒共配备有4个相机接口。

新增的入射光路接口，有助于整合额外的光源和激光系统。如Infinity 光操作系统、Infinity TIRF、FRAP、光转换等先进显微技术所需激发光源，实现FRAP、光活化、激光消融、激光镊等新型显微技术应用。

 

3、快速荧光成像高速图像采集领域

对于高速成像，徕卡DMi8集成外部荧光转换功能(External Filter Wheel, EFW)、自动荧光照明强度管理系统(Fluorescence Intensity Manager, FIM)，微秒快速LED光源，使得DMi8 sequencer可以在微秒级别速度和精度下同时控制，实现快速、准确地荧光成像。有助于捕捉活细胞的动态过程，将光毒性以及光漂白降至最低。

利用LAS X 和Dual-View 图像分割器，同时获取多通道图像。

专用FURA2成像系统，物镜对340nm处的UV透射高达60％，配合极快的EFW转换功能（24ms切换时间），足以捕捉如活细胞内钙离子流量快速动态测定过程。

 

5、宽视场成像

DMi8标配FOV 19 mm视场摄像头端口、大幅面 sCMOS 相机，极大拓展了图像视野。

 

4、自适应调焦控制

利用 LED 光束辅助功能的徕卡自适应调焦控制(Adaptive Focus Control, AFC)和闭环聚焦（closed-loop focus）调焦控制系统，只需在Leica LAS X软件单击一次激活自动对焦以及闭环调焦按钮，Leica DMi8 调焦驱动器以 20 nm 的重定位精度实现自动实现精确地保持焦平面。即使在活细胞延时实验（timelapse experiments）中因实验体系出现剧烈温度变化、载物台 x-y 移动，物镜焦平面也始终保持稳定。AFC完全集成到 LAS X软件中，并可与基于图像的自动对焦系统结合使用，典型应用是活细胞成像、孔板筛选、多位置实验和比率成像。

 

6、模块化配置

DMi8 采用模块化设计，从手动到电动各种组件可自由组合，以适应不同的透射光方法和荧光应用方法。相应编码组件或电动组件的设置状态可自动从显微镜传输到连接的PC（USB）上的软件，作为图像信息要素被记录保存。

组成配置

Leica DMi8A、DMi8C、DMi8M三款倒置显微镜中，Leica DMi8A的透射光轴(Transmitted-Light Axis)、入射荧光轴（Incident Light Axis）、光学载体(Optic Carrier)、荧光滤块转盘（Reflector Turret）、物镜转盘（Nosepiece）、载物台即可用配置全电动工作组件，也兼容带编码的各种工作组件。DMi8C则适用于装配编码型器件工作，而DMi8M则只能配置全手动控制型组件。

细胞生物学实验中，因观察对比方法、研究手段的不同，入射照明光源、入射光轴、光学载体元件、荧光滤块、物镜配置、图像光路出口布局、载物台及外围控制设备配置要求不同。徕卡为电动、编码、手动主机分别提供了具有不同无限远光学成像输出口（camera ports）设置的基座(Base Stand)。主体基座根据应用需求，需添加入射光轴（incident light axis）、透射光臂(transmitted light arms)、LED透射光源、聚光镜(Condensers)套件、DIC/相差/IMC/前端模块(front modules)/放大倍数转换器(magnification changers)、荧光照明光源、荧光滤片立方模块、物镜调焦控制、物镜转盘、载物台及驱动控件、目镜筒、数码摄像头等搭建相应的应用配置系统。

除电动型DMi8A主机，需配置电控制元件（Control-Elements）。DMi8C型主机还需在机身正面根据编码元件配置种类，增配相应LED指示和控制按钮。

Leica Assistant LAS Store & Recall Tube（12730488），即Leica LAS Store and Recall Module，徕卡显微镜助手、徕卡LAS软件存储和调用功能模块，会自动将生成每张图所设定的照明强度和对比度方法、放大倍率和曝光时间等设置直接与图像一起存储。只需在图像上单击一次鼠标，可快速恢复所有显微镜设置，确保相同实验中所有所获取图像成像条件完全相同。便于工业和生命科学应用中重新检查比较样品，无需在实验室中通过手工记录和人工记录这些显微镜设置要素。

Leica DMi8系列倒置荧光显微镜，具有开放型架构，视具体应用需求不同，各功能模块采用积木式组合配置。
DMi8A采用电动工作组件，可通过机身正面触摸屏或联机Leica Application Suite X应用软件，镜头聚焦、光路转换、光源强度调整、快门关闭、物镜及光学元件的转入/转出光路全自动完成，操作简化，并可减少人为操作失误。同时，系统自动将成像所涉及光学元件、光路设置信息整合到所生成图像文件中，用户可随时查看和调用图像生成所需的各项设置、条件参数信息，确保可在相同条件下获得高质量细胞图像。
DMi8A的具有左、右、上、底部四路光学图像输出口，数码相机既可以采用三目镜筒安装，也可以安装与机身左、右侧。系统支持多相机组合应用方案，分别为不同检测通路分配专用图像采集光路。系统可根据预先设置，实现多路图像信号的实时同步采集。
对于Fura2、FRET和光转化等多光路、同步荧光图像采集分析应用，为实现不同荧光检测通道信号的快速切换，DMi8A可引入电动控制的Leica EL6000光源及快速滤光片轮（External Filter Wheel, EFW)相结合,配合专用滤光片模块，可实现24ms完成荧光通道的超快速切换，实现高达40fps的图像采集传输性能。
利用DMi8具有的双Infinity-Port照明光口配置（对应于T-house 3），DMi8A可添加TIRF功能所需的激发光源模块及TRIF专用物镜，构建TRIF分析平台。
