三、HTRF检测应用中多功能荧光酶标仪的选型要略
常规FI、TRF、FRET检测,人们可能只关注到荧光酶标仪的激发、发射波长配置与测试要求契合的问题。而当了解HTRF标准流程后,对优化荧光检测条件的意义和其中涉及的技术要素之复杂,兴许会有感悟。
开展HTRF测试前,实验者首先须确认是否有可利用的HTRF认证多功能荧光酶标仪资源。

确认多功能荧光酶标仪属HTRF认证型号后,还应核实是否配置有HRTF检测规定的专用光学元件。不同品牌机型,制造商提供的HTRF功能组件有区别。
如VantaStar、Folarstar Omega、SpectraMax iD5、Synergy NEO2均须添加HTRF专用的具有Z轴调节功能的增强型时间分辨荧光模块(Enhanced TRF module)、320/340nm紫外激发滤片、520/620/665nm发射荧光滤镜这三件套。
相对而言,MD SpectraMax i3、SpectraMax Paradigm、SpectraMax i3x这三款荧光酶标仪,由于采用的是检测功能模块化设计,必要时直接将选配的HTRF检测卡盒插入主机卡槽就可以启动测试。实验者无须为激发与发射滤光片的波长与带宽、二向分色镜截止波长范围这类问题纠结。

拥有HTRF认证资质的荧光酶标仪、配套光学元件支持,再加上HTRF检测试剂,开展HTRF技术研究方可谓万事俱备、水到渠成。
四、HTRF检测程序与同类TR-FRET方法间的兼容性
目前,同样是基于TR-FRET技术开发的检测试剂, HTRF之外还有MD IMAP Assays、BellBrook Transcreener assay 、Invitrogen LanthaScreen TR-FRET Kinase Assay、Bioauxilium Thunder Enhanced TR-FRET assay、PE Lance TR-FRET Assays等。
单论测试光谱数据,这些TR-FRET试剂的检测光谱设置基本是通用的。那实际工作中,其他同类产品能否用HTRF的检测标准程序测定?
分析结果是:不行。

PE-Cisbio强调的是,HTRF认证测试流程、方法的应用范围严格限于HTRF试剂和技术使用,不适用于用于其他TR-FRET产品的测定。
TRF-FRET检测具有时间高度灵敏的特性。在各种不同来源样品测试条件下,不同TRF-FRET试剂盒的荧光光谱稳定性、半衰期特征的差异,注定了检测过程的积分时间、脉冲数量、时间延迟几个关键参数难以完全匹配。尽管两种试剂盒测定的激发波长、发射波长峰值特征一致,但不代表TRF-FRET过程中供体、受体两种荧光信号的动力曲线相同。
HTRF试剂采用有大环结构的穴状态镧系配合物,其光谱稳定性优异,与特殊优化的缓冲液配合,可以实现0.6-1ms的超长荧光寿命。其他的TR-FRET技术中所用的镧系配合物、FRET能量受体释放的FRET荧光信号动力学特征除非能高度还原HTRF产品FRET过程的荧光表现。否则,在同一型号荧光酶标仪上用HTRF提供的标准测试程序测定其他厂商试剂,难免弄巧成拙、东施效颦。

对比Ciosbio HTRF-Eu/red Assay、LanthaScreen Eu Kinase Activity Assay在Perkin Elmer EnVision多功能荧光酶标仪上检测所用的测试参数,可发现:尽管两个试剂盒规定的激发波长、双荧光发射波长设置基本一致(测试年份不同,同一型号仪器出厂预装滤光片规格略有变动),但脉冲信号的积分时间延迟(HTRF-60µs VS LanthaScreen-100µs)、积分时间长度(HTRF-400µs VS LanthaScreen-200µs)的设定迥异。
这证实了我们分析的结论:不同TR-FRET试剂盒在荧光多功能酶标仪上的规定的测试条件设置存在较大区别,其标准检测程序相互难以有效兼容。
表5的信息还表明:与Ciosbio HTRF-Eu比,LanthaScreen Europium激发的背景荧光衰减慢、而FRET供体和受体的荧光寿命短;LanthaScreen Eu检测时间延迟拉长,极大地限制了检测积分窗口的选择余地和测试灵敏度的提升空间。
